Figure 5. — Akt is involved in COM crystals-induced tight junction disruption by activating ASK1 and p38. (a) Akt inhibitor MK-2206 alleviated the down-regulation of ZO-1, occludin, and the activation of ASK1 and p38 induced by COM crystals. Protein levels were detected by Western blot and the band intensities were analyzed by Image-Pro Plus 6.0. (b) Akt inhibitor MK-2206 inhibited the apoptosis induced by COM crystals. MDCK cells were pretreated with or without 5 μM MK-2206 for 24 h, then were incubated with 1 mM COM crystals for 48 h. Cell apoptosis was analyzed using flow cytometry by Annexin V-PI double staining. Representative data from repeated experiments performed in triplicate were presented on the right panel. **p < .01 versus control; ##p < .01 versus COM.
