Luciferase reporter constructs were generated by cloning the 3′UTR of LIFR, ATG16L1, TGFBR and IL1RAP downstream of the renilla luciferase in psiCHECK2. (A–D) Top panel shows the sequence alignment of the predicted miR-H1 (in red) binding site on the target and gene (in black). HEK293 and HOK cells were transiently co-transfected with reporter construct and either miR-H1 or control mimics. Constitutively expressed firefly luciferase readings were used for normalization. Histograms shows impact of miR-H1 on normalized renilla activity compared to experimental controls. Error bars represent mean ± SD from three biological replicates.
