CH-5 decreased transcriptional levels and protease activity of MMP-2 in HGC-27 cells. (A) HGC-27 cells were treated with 0, 10, or 40 µM of CH-5; after 24 h the total RNA was isolated and then subjected to conventional RT-PCR using primers for MMP-2 and housekeeping gene GAPDH. A representative gel (left) and densitometry analysis from three independent experiments (right) are shown; (B) HGC-27 cells were treated with CH-5 (0, 10, 20, and 40 µM) for 24 h and then the activity of secreted MMP-2 was measured by gelatin zymography assay.
