Secretion of IL-6 by the peri-pancreatic adipose tissue from obese mice is synergistically increased in the presence of natural killer (NK) cells. Purified NK cells from splenocytes and adipocytes from the peri-pancreatic fat were obtained from WT mice on CD or KC mice on HFCD as described in Section “Materials and Methods.” NK cells treated with IL-2 (10,000 U/ml) in the presence and absence of autologous adipose cells at (NK:adipose cells; 1:1) were cultured and supernatants were harvested after 7 days and the secretion of IL-6 (A) and IFN-γ (B) were determined. One of the three representative experiments is shown in the figures. Cytotoxicity of NK cells cultured as described in (A,B) were assessed against ST63 tumor cells using the standard 4-h 51Chromium release assay. The LUs 30/106 cells were determined as described in Figure 1B (C). One of three representative experiments is shown in the figure.
