Double-mutant analysis of ply2 and smax1. (A) Phytochrome B (phyB)-dependent germination of the wild-type (WT, Col), ply2, smax1-3, and ply2smax1-3 double-mutant. After treatment with far-red light for 10 min, the seeds were then either irradiated (phyBon) or not (phyBoff) with red light for 10 min, and then kept in darkness for 4 d. Mean values (±SD) of germination rates were determined from three experimental replicates with at least 50 seeds per plate. (B) Hypocotyl elongation assay. The images show representative 6-d-old seedlings of each genotype grown on half-strength MS-SUC (0.5%) media under short-day conditions (10/14 h light/dark); scale bar, 5 mm.
