Effect of antibiotics and antibiofilm peptides on biofilm growth evaluated under both inhibition and eradication conditions. (A) P. aeruginosa PAO1 biofilms were grown in BM2 glucose minimal medium (B) S. aureus MRSA biofilms were grown in 10% TSB supplemented with 0.1% glucose. Inhibition is shown in the upper panels and eradication assays in the lower panels. The adhered biomass within the wells was quantified by CV staining (purple lines) while the metabolic activity in the preformed biofilm samples was quantified by conversion of TTC dye to the red formazan end product (red lines). The vertical gray bars indicate the concentration of antibiotic or peptide that resulted in greater than 90% reduction in bacterial planktonic growth as measured by recording the OD600 in each well prior to rinsing the spent media. Data represents the average ± standard deviation (SD) of at least three biological replicates.
