The Hypomorphic Apoeh/h allele: The wildtype mouse Apoe allele is composed of 4 exons on chromosome 7. A neomycin resistance cassette flanked by loxP sites introduced into intron 3 reduces ApoE expression levels of the gene in all tissues, presumably by interrupting RNA splicing. Cre-mediated removal of neo cassette restores Apoe expression to physiological levels.
