Concept of the Candida-reactive lymphocyte assay. Peripheral blood mononuclear cells (PBMC) are co-incubated with Candida spp. specific lysate (e.g., C. albicans) and stimulated with CD28 and CD40 antibodies. T cells react to the presented fungal peptides on the surface of antigen presenting cells (APC) and upregulate the activation markers CD69 and CD154 (CD40L), which are quantified by flow cytometry after 5 h stimulation at 37°C and 5% CO2.
