FH and FB are increased specifically in DENV antigen-positive HUVEC and MDM. (A, B) HUVEC (A) and MDM (B) were isolated and were left uninfected (UI) or DENV infected. At 48 hpi, cells were fixed and immunostained for DENV (green), FH (orange), and FB (red). Nuclei were stained with Hoechst (blue). Cells were imaged with an Operetta high-content imaging system at a ×20 magnification. (C, D) Images of 49 fields of view were analyzed, and the intensity means were calculated using Harmony software. The intensity means for FH and FB were compared in uninfected (gray bars) versus DENV-infected (black bars) wells and in DENV-negative cells (gray bars with a hatching pattern) versus DENV-positive cells (black bars with a hatching pattern) within a well. HUVEC (C) and MDM (D) were analyzed. Results represent the mean ± standard deviation for triplicate samples and are representative of those from three independent infection experiments *, P < 0.05, one-way ANOVA/Tukey's test.
