Figure 5

Effect of PL on phosphorylation of cyclic AMP response element-binding protein (CREB) in Aβ25–35 plaque-stimulated HT22 cells and mice. Cells were treated with PL (100 μg/mL) for 1 h and incubated with Aβ (10 μmol/L) for a further 23 h. Cell lysates were assessed by Western blot using pCREB antibody. (A) Quantification of pCREB protein was normalized to β-actin. After stereotaxic injection of Aβ25–35 plaque, mice were treated with vehicle or PL for 17 days; (B) pCREB expression was explored in the dentate gyrus (DG). Scale bar = 100 μm. Values are mean ± standard error of the mean. * p < 0.05 vs. control group; # p < 0.05 vs. Aβ25–35 plaque group. The arrowheads in magnified images point to pCREB/4’,6-diamidino-2-phenylindole (DAPI) double-labeled neurons.
