Internalization. Human breast cancer cells MDA-MB-231 were incubated with Cy 5.5-labeled Dox-loaded PC liposomes. After 1 and 3 h, PC-liposomes were removed, and the endolysosomes were stained by using LysoTracker Red DND-99. The cells were fixed in 4% paraformaldehyde solution and mounted by using DAPI-contained mounting medium. The fluorescence of the Cy 5.5-labelled Dox-loaded PC liposomes and MDA-MB-231 cells were observed by using confocal laser scanning microscopy (CLSM). The fluorescence of the Cy 5.5 and the anticancer drug Dox were excited, respectively, at 633 and 488 nm, and their fluorescences were observed at 650 and 520 nm, and shown in grey and green. The nucleus and endolysosomes of the MDA-MB-231 cells were marked and observed by using DAPI and 577/590 nm (excitation/emission) detecting conditions, respectively, shown in blue and red.
