Schematic illustration of the procedure for single-molecule detection of DNA hybridization. (A) Evanescent field generated on the glass surface by the excitation ray (532 nm) contributes to a single-molecule detection during hybridization between Cy3-labeled DNA and biotinylated DNA immobilized on the glass plate. Fluorescence is transmitted through the objective lens, which is detected by the CCD camera. (B) A hybridization event is indicated by a bright spot on the CCD camera images.
