Silencing PRMT5 decreases PI3K/AKT signaling. A, Western blot analysis of PRMT5 in AML12 cells following lentiviral shRNA knockdown of PRMT5. B, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis of differentially expressed proteins with PRMT5 knockdown. Total protein extracted from scramble or PRMT5 knockdown cells was analyzed for the expression of key proteins involved in more than 20 signaling pathways. Signals were normalized to α- and β-tubulin. Results were obtained from two independent experiments performed in duplicate. C and D, reduction of phospho-AKT Thr-308 and Ser-473 was observed in PRMT5 knockdown liver cells (C) as well as PRMT5 knockout MEF cells (D). KO, knockout. E, a high-fat diet up-regulated the hepatic expression of phospho-AKT Thr-308 and Ser-473.
