CHE protein stability is controlled by the proteasome. A cell-free degradation assay was performed on 35S::CHE-GFP grown in LD cycles. Samples were collected at ZT8 (light) or ZT16 (dark), and protein was extracted at 4°C and split into tubes at room temperature for mock treatment or treatment with 1 µm MG132 for the indicated times. Samples were normalized to time 0 for quantification. Error bars represent sd of three biological replicates. WT, Wild type.
