PDZ1 (PDB ID: 1ihj19) is inserted into a binding-deficient fusion-competent Sindbis virus E2 envelope protein and used to pseudotype lentivirus. The CTP (e.g. DARPin, PDB ID: 4j7w35) is fused to a short pentapeptide ligand (TEFCA). Mixing of these two components enables covalent functionalization of the lentivirus with the CTP through the formation of a disulfide bond between the PDZ1 and TEFCA ligand.
