TADs boundaries are insulated by CTCF/cohesin complex (C). Within TADs, enhancer and promoter form chromatin loops with super-enhancers and their target genes tend to have better phase separation (depicted in dark grey) due to the higher concentration of local TFs and transcriptional machinery than typical enhancers (depicted in light grey) (C).
