Ca+2 and PKC regulation of AC isoforms in the HEK-ACΔ3/6 cell line. (A) HEK-ACΔ3/6 cells were transiently transfected with the Venus control plasmid, AC1, AC2, or AC8. Cells were incubated with 10 μM A23187, 10 μM PGE2, or a combination of both, as indicated, and cAMP accumulation was measured. (B) Venus, AC1, AC2, or AC7-transfected HEK-ACΔ3/6 cells were incubated with 1 μM PMA, 50 μM forskolin, or a combination of both, as indicated, and cAMP accumulation was measured. Data represent the mean and S.E.M. of three to four independent experiments conducted in duplicate. Statistical analysis was performed using one-way ANOVA, followed by a Dunnett’s comparison. *P < 0.05; **P < 0.01; ***P < 0.001; ****P < 0.0001 compared AC-transfected cAMP responses to the responses of the Venus-transfected cells for each stimulation condition.
