Mixture of human Hsp90α with Hsp90β yields little heterodimer formation. Equal amounts of purified human Hsp90α and His-tagged human Hsp90β (His₆-V5-Hsp90β) were incubated for the indicated times at 30°C under the same conditions used for GR•Hsp90 heterocomplex assembly. His₆-V5-Hsp90β was adsorbed to a nickel matrix and Hsp90α and Hsp90β were immunoblotted and scanned. The amount of Hsp90α in a nickel pellet prepared from a control sample with hHsp90α and hHsp90β without His-tagged Hsp90β was the same as the 0 time pellet prepared from the mixture.
