Role of EEA1 in CXCL12-instigated Akt phosphorylation.
A, CXCL12-instigated Akt phosphorylation was examined in cells depleted of EEA1. Serum-starved HeLa cells transfected with control siRNA (siLuc) or two distinct siRNAs against EEA1 (siEEA1-1 and siEEA1-2) were treated with 10 nm CXCL12 for 5 min at 37 °C. Whole-cell lysates were analyzed by immunoblotting for the phosphorylated or total forms of the indicated proteins. B–D, immunoblots were analyzed by densitometry. Bars represent the mean relative levels of pAkt-Ser473 (B), pFoxO1/3a (C), or pTSC2, pGSK3β, and pERK-1/2 (D) to siLuc- and CXCL12-treated samples. Error bars represent the S.D. Data were analyzed by two-way ANOVA followed by Tukey's multiple comparison test. p values are indicated.
