FIGURE 3

Aβ treatment decreases length of neuronal processes. Neurons from n = 3 differentiation experiments were treated with either Aβ25-35 peptides or Aβ1-42 oligomers at a range of concentrations (0–20 μM) and stained with the live cell dye calcein acetoxymethyl (Calcein AM). Neurite length was measured by automated imaging software and unpaired t-tests compared neurite length in treated cells to untreated controls. (A) Representative images of neurons treated with Aβ1-42 oligomers at 0 μM, 1.25 μM, and 2.5 μM for 48 h. (B) Representative images of neurons treated with Aβ25-35 peptides at 0 μM, 6 μM, and 20 μM for 48 h. (∗∗p < 0.005, ∗∗∗p < 0.001). Scale bars represent 50 μm. (C) Characterization of Aβ1-42 oligomers with analytical ultracentrifugation. Ultracentrifugation and western blotting shows sizes of proteins present in Aβ1-42 sample preparations.
