PRRSV induces both eIF2α phosphorylation-dependent and -independent host translation shutoff. (A) MARC-145 cells grown in six-well plates were treated with 0.5 mM Ars at 37°C for 1 h. The medium was changed to medium containing different concentrations of ISRIB, and after 25 min, the cells were treated with puromycin, incubated at 37°C for 25 min, and lysed for Western blotting. (B) MARC-145 cells grown in six-well plates were infected with PRRSV (MOI = 0.5). At 36 h postinfection, the cells were treated with 20 μM ISRIB for 25 min and with puromycin for 25 min. Western blotting was performed to analyze the levels of translation and eIF2α phosphorylation.
