Xrn1 expression levels decrease after autophagy induction. (A) WT (WLY176) cells were grown to mid-log phase in YPD (+N) and then nitrogen starved (-N) for 1 h. Total RNA was extracted and RT-qPCR was performed. Results shown are relative to the level of XRN1 mRNA in rich conditions (+N), which was set to 1. TFC1 was used to quantify relative expression levels. Results shown are the mean of 6 independent experiments +/− SD. (B) Cells (EDA161) endogenously expressing Xrn1-3xHA were grown to mid-log phase in YPD (+N) and then nitrogen starved (-N) for 1 h. Protein extracts were analyzed by SDS-PAGE and blotted with anti-HA antibody or anti-Pgk1 (loading control) antiserum. (C) Densitometry of n=4 blots represented in B.
