Knockdown or inhibition of USP11 reduces PEG10 mRNA and protein levels. (A–D) A549 cells (A, B) or Beas2B cells (C, D) were transfected with control shRNA or Usp11 shRNA for 72 h. (A and C) RNA was extracted and then analyzed by qRT-PCR with Usp11 and Peg10 primers. The relative expressions of Usp11 and Peg10 were normalized to Cont shRNA. (B and D) Cell lysates were analyzed by immunoblotting with USP11, PEG10, and β-actin antibodies. PEG10 protein levels were quantified by ImageJ software. (E–H) A549 cells (E, F) or Beas2B cells (G, H) were transfected with empty vector plasmid, V5-tagged USP11, or V5-tagged USP11 mutant C318S (Usp11c318s-v5) plasmid for 48 h. Peg10 mRNA expression (E, G) and PEG10 protein levels (F, H) were determined. Shown are representative blots from at least three independent experiments.
