(i) The resting integrin α4β7 with closed conformation binds to MAdCAM-1 and VCAM-1 in low affinity. CXCL10 (ii) and CCL25 (iii) induce two different intermediate open conformers of α4β7, which show selective binding to VCAM-1 or MAdCAM-1, respectively. (iv) Mn²⁺-induced fully open conformer of α4β7 shows nonselective high-affinity binding to both ligands.
