A Specific Polyplex Ratio Achieves Optimal Gene Expression and Biocompatibility in Primary Astrocytes

An AnPn analog S2 A5P10 or polyethylenimine (PEI) was mixed with 2.5 μg/mL luciferase plasmid in different w/w ratios (1:1, 2:1, 4:1, and 8:1). Astrocytes (150,000/well) were treated with polyplexes for 3 hr. The polyplex-containing media was removed, and fresh culture medium was added. The (A) luciferase, (B) lactate dehydrogenase (LDH), and (C) metabolic activities were measured 48 hr post-treatment. Data represent mean ± SEM for two donors with a minimum of triplicate determinations/donor (*p < 0.05, **p < 0.01, and ***p < 0.001 for 1:1 w/w versus other ratios for S2 A5P10 or PEI, #p < 0.05, ##p < 0.01 for S2 A5P10 versus PEI at same w/w).
