SIN inhibits proliferation and promotes apoptosis of B16-F10 cells.

Notes: B16-F10 cells were treated with SIN (0, 12.5, 25, 50, and 100 μM) for 24 hours. (A) The cell viability was analyzed by CCK8 assay. (B) The cell apoptosis was analyzed by Annexin V–PI flow cytometry. (C) Apoptotic cell quantification for three independent experiments. (D) The protein level of Bax and Bcl-2 was detected by Western blot. (E) Quantification of Bax/Bcl-2 in Figure 1D. (F) The activity of caspase-3 was measured by caspase-3 ELISA kit. All the experiments were repeated at least three times. β-actin was used as loading control. *P<0.05, **P<0.01 versus untreated control.
