Analysis of transcription factors by protein/DNA array. (A) Clustering analysis based on 345 TFs was undertaken in the KD and NC groups of RL-952, AN3-CA, HEC-1A, and HEC-1B. (B) Reading from the images of protein/DNA arrays, the plots of different TFs show the relative activity-level clusters of the NC and KD groups in RL-952, AN3-CA, HEC-1A, and HEC-1B. (C) Analyzed by MeV software, clustering analysis based on 345 TFs-chips between the NC and KD group revealed that the 20 TFs show the same change trend of transcriptional activities in the four RL-952, AN3-CA, HEC-1A, and HEC-1B EC cells. (D) The function of different TFs may be mediated by ERRα in the cellular nucleus as presented by the schematic diagram. Downregulation of ERRα then induced inhibition of TFEB which was mediated by PGC-1α and involved the mTOR signaling pathway. In addition, mediated by TCF, the downregulation of ERRα could cause Oct3/4 upregulation and participated in the Wnt signaling pathway.
