At E12.0 muscle, nerve, vasculature, and PPF-derived connective tissue and tendon progenitors are present in developing diaphragm region. (A–C, E, G–R) Into PPFs (as shown by GFP+ Prx1-derivatives in Prx1CreTg/+;Rosa26mTmG/+), muscle progenitors (Pax7+MyoD+ via immunofluorescence) migrate from the somites and phrenic nerve axons (Neurofilament+ via immunofluorescence) extend from the neural tube. Muscle progenitors and phrenic axon tip completely surrounded by PPFs.
