Antiallodynic effects of FTY720 do not undergo tolerance. Mice were injected (i.p.) with 6 mg/kg FTY720 or vehicle once daily for 6 days, followed by 6 mg/kg FTY720 on day 7, and assessed for paw withdrawal threshold (mechanical allodynia) or acetone-induced paw flinching (cold allodynia) on days 1 and 7, as illustrated in the schematic timeline (A). FTY720 produced significant reductions in both mechanical allodynia (B) and cold allodynia (C) on days 1 and 7 compared with responses in control mice receiving vehicle on day 1.
