PDX1 is transcriptionally regulated by SIRT5. MIN6 and INS-1 cells were an empty plasmid vector, a plasmid vector containing SIRT5, SCR, siSIRT5 or SIRT5 (T69A). Following 48 h transfection, the (A) mRNA and (B) protein levels of PDX1 was detected using reverse transcription quantitative polymerase chain reaction and western blotting analyses, respectively. (C) SIRT5 was knocked down in treated MIN6 and INS-1 cells, and anti-H4K16ac antibodies were used to perform qChIP assays. Each experiment was repeated three times. The results are presented as the mean ± standard deviation. *P<0.05 vs. SCR and #P<0.05 vs. vector. SIRT5, sirtuin 5; Vector, pcDNA3.1 plasmid vector; SCR, scrambled small interfering RNA; siSIRT5, small interfering RNA targeting sirtuin 5; PDX1, pancreatic and duodenal homeobox 1; qChIP, quantitative chromatin immunoprecipitation.
