Granzyme B activity can be distinguished from caspase-8 activity. HeLa cells overexpressing CD95 were transfected with (A) NES-ELQTD-mGFP (for caspase-8), and NES-RIEADS-mCherry for granzyme B or (B) NES-ELQTD-mGFP for caspase-8 and NES-VGPD-mCherry for granzyme B and imaged by confocal microscopy upon incubation with 1 µg/ml IZsCD95L.
