HET0016 and TRPV1 inhibition reduce ROS production. (A, B) TRPV1 knockout (KO), A784168 (A7), and HET0016 each attenuated the ROS production induced by 20-HETE mimetic 20-5,14-HEDGE (HEDGE, A) or 1-h OGD (B), as measured by the presence of 2',7'-dichlorodihydrofluorescein diacetate (DCFDA). The combination of A784168 and HET0016 did not achieve additive effects. (C) Western blot analysis revealed increased carbonyl immunoreactivity, indicative of H-I-induced oxidative stress, in the ipsilateral hemisphere at 3 h of recovery. HET0016 and A784168 each reduced carbonyl immunoreactivity. The combination of A784168 and HET0016 did not achieve additive effects (n=4-7/group).
