Schematic of the FNA-based approach for probing oligomers. (A) The scheme showing the peptide-conjugated FNA construct. The polymer had a thiol group and biotin (red blob) at the terminals; also, three DBCO groups were located at specified distances. Three Aβ (14–23) molecules were attached using a metal-free click reaction. The distances are shown according to the experimental design. (B) Force spectroscopy set up. The AFM tip was functionalized with an FNA construct and the surface with streptavidin. The Aβ(14–23) trimer is formed by association of three monomers. Note, schemes are not to scale.
