Previous approaches for i to i+4 stapling include (a) lactamization, (b) thioether bridging, (c) perflurobenzene SNAr, (d) alkene RCM, and (e) CuAAC “Click” chemistry. Present work, (f) applies the Glaser reaction to form a rigid, linear 7 Å diynyl linker between i to i+4 spaced propargyl serine residues.
