TRAIL sensitization by garcinol is independent of ROS levels. (A) For measurement of ROS production, cells were treated with 2 μM garcinol for 2 h and then stained with H2DCF-DA dye. Fluorescence was detected by fluorescence microscopy and flow cytometry; (B) After pretreatment with the indicated concentrations of NAC, GEE and trolox for 30 min, cells were stimulated with 2 μM garcinol plus 50 ng/mL TRAIL for 24 h. Levels of apoptosis were assessed by flow cytometry, and western blot showing the PARP, DR5, c-FLIP and actin.
