Figure 3

Infusion of IL-22Ab suppressed cell apoptosis, increased the expression of Foxp3, and reduced cytokine expression in vivo. (a) At 14 days after cell transfer, Fas expression in host B cells and FasL expression in donor CD8+ T cells from the spleen were examined by flow cytometry. (b) aGVHD was induced as previously described. After 14 days, mice were sacrificed, and the spleen cells were stained for forkhead box p3 (Foxp3), CD4, and CD8. Representative contour plots of CD4+Foxp3+ and CD8+Foxp3+ cells determined by flow cytometry. Data are shown as the means from five independent experiments. Each group included five mice. (c, d) Representative plots of percentages of CD4+ T cells expressing various cytokines, including IL-17, IL-4, interferon- (IFN-) γ, and tumor necrosis factor- (TNF-) α, as determined by flow cytometry. Bars show the mean ± SEM of six independent experiments. ∗∗ p < 0.01.
