SAMHD1 expression negatively correlates with myeloblast levels in AML patient-derived PBMCs. (a) Five million PBMCs from 22 AML patients were collected and analyzed via immunoblotting for endogenous SAMHD1 protein. PBMCs from 2 ‘Normal’ (healthy) donors and THP-1 cells were used as a reference. GAPDH was a loading control. The percentages of blasts (%) in patient PBMCs are shown. Relative SAMHD1 levels were calculated by densitometry analysis. (b) Non-parametric correlation (Spearman) analysis between relative SAMHD1 levels and blast percentages (P = 0.046, R2 = 0.1935). Patient #1 sample was excluded from analysis due to low protein concentration of the cell lysate.
