The qRT-PCR analysis demonstrated that the upregulation of miR-139-5p resulted in a reduction of Notch1, N-cadherin, Vimentin and Fibronectin mRNA expression and an elevation of E-cadherin mRNA expression. (g and h) The Western blot analysis showed that the protein expression of Notch1, N-cadherin, Vimentin and Fibronectin decreased, and the protein expression of E-cadherin increased in LN229 and U87 cells after transfection with miR-139-5p. (i) Immunofluorescence showed miR-139-5p overexpression increased E-cadherin expression and decreased Vimentin expression in glioma cells. This experiment was repeated three times.
