Secondary structure of the 5′ TAR element of HIV-1BH10 and synthetic oligonucleotides used in strand transfer assays. The 5′ and 3′ ends of donor and acceptor molecules are indicated in the TAR structure. The bottom panel shows a standard primer–donor–acceptor complex with expected donor extension (DE) and strand transfer products (TP) (added nucleotides are shown in blue). The R33B/20A hybrid was also used in template-primer binding affinity and RNase H cleavage assays and for the determination of dissociation rate constants.
