Representative whole-cell recordings of TASK-3 (A,B, n = 6) and TASK-3/2Loop2 (D,E, n = 6) K+-mediated currents recorded according to the voltage protocol described in the legend of Figure 1. The results in (AC) correspond to TASK-3 WT channels and those in (DF) to TASK-3/2loop2 construct. In (A,D), the extracellular medium was 135 mM Na+ and 5 mM K+. In (B,E), the extracellular Na+ was replaced by an equimolar amount of K+(140 mM). (C,F) shows the representative current-voltage relations for TASK-3 and 2loop2 channels under different extracellular K+ concentrations (closed circles 5 mM; open circles 140 mM).
