Effect of AVV2/8-DYRK1A gene transfer on brain calpain activity. A) Calpain activity, B) calpastatin and C) calpain 2 domain I protein levels in brain of Cbs+/- mice with intermediate hyperhomocysteinemia (hhcy) with (hhcy/AAV) or without (hhcy) AAV2/8-DYRK1A treatment, and control (CTL) mice. Calpain activity was determined by measuring the proteolysis of the substrate for 21 min. Calpastatin and calpain 2 domain I protein values were obtained by normalization of images from calpastatin and calpain 2 domain I to total proteins marked with Ponceau-S. Data were normalized to the mean of CTL mice. The results are represented as means ± SEM. n = number of mice.
