Enriched Thy1.1⁺ OT-I T cells (~10⁶) from naïve Cblb^+/+ or Cblb^−/− mice were adoptively transferred into naïve Thy1.2⁺ Cblb^+/+ or Cblb^−/− mice. On the following day, recipients were vaccinated with strain #55 yeast engineered to express the OT-I epitope (10⁶ CFUs). On day 22, dLNs were harvested to enumerate cytokine-producing CD8⁺ T cells.
