Two HPV- HNC cell lines (UM-SCC4 and UM-SCC19) and two HPV+ HNC cell line (UM-SCC47 and UM-SCC104) were stimulated with 1μg/mL rituximab or 1μg/mL cetuximab as indicated. After 48h, the cells were treated with 50IU/mL IFNγ and 30ng/mL TNFα. (A)After 24h, the expression levels of the genes indicated were determined by RT-qPCR. Gene expression was normalized against GAPDH mRNA levels. Similar results were observed in two independent experiments. (B) After 48h the concentration of CXCL9 was determined by Enzyme-linked immunosorbent assay in supernatants of three different experiments. (C) The supernatant isolated after 48h were used to study CD14-depleted PBMC migration in trans well assays. P value were determined by unpaired t-tests.
