Effects of hindsiipropane B on NF-κB and AP-1 signaling cascades in HIV-1 Tat-stimulated CRT-MG cells. Cells were pretreated with hindsiipropane B for 1 h and then stimulated with 50 nM HIV-1 Tat for 1 h. Whole cell or nuclear extracts were collected and assayed for p65 and IκBα (A) or c-Jun and c-Fos (C) by Western blotting using relevant antibodies. (A) Cell lysates were collected. The amount of phosphorylated p65 and IκBα in the cell lysates, as well as p65 in the nuclear extracts, was measured by Western blotting. (B, D) Nuclear extracts from HIV-1 Tat-stimulated cells were collected and assayed for DNA binding activity of NF-κB and AP-1 by EMSA.
