Proteasome activity regulates hIAPP turnover in a lysosome-independent manner. Western blot analysis of intracellular hIAPP turnover in proteasome and lysosome impaired cells was performed. Human islet cells were cultured in 20 mm glucose containing media for 5 days, followed by additions of LC (5 μm), pepstatin (5 μm), or both for additional 12 h. Histogram depicts mean ± S.E. changes in hIAPP protein levels for three independent experiments.
