15d-PGJ2-loaded NC reduce MSU-induced IL-1β maturation in vitro in a PPAR-γ-sensitive manner. LPS-primed BMDMs were treated with 15d-PGJ2-loaded NC 0.03, 0.3, or 3 μM; or with free 15d-PGJ2 (A), or with 15d-PGJ2-loaded NC + GW9662 (10, 1 and 0.1 μm) (B) 30 min before MSU stimulation. Supernatants were collected 5 h after MSU stimulation and IL-1β levels were quantitated by ELISA. Results are mean ± SEM, n = 6 wells per group in each experiment, two independent experiments (*p < 0.05 vs. control group; #p < 0.05 vs. vehicle mg/kg group, **p < 0.05 vs 15d-PGJ2-loaded NC.
