Disruption of L12–L10 protein interaction by IMB-84 and IMB-87. (A) Demonstration of L10–L12 interaction by GST pull-down assay. Purified GST-L12 or GST proteins were bound to glutathione-sepharose beads, and then incubated with purified His-tagged L10. Here show protein bands of resulting pull-down products after western blotting with anti-GST and anti-His antibodies. (B) and (C) Compounds IMB-84 and IMB-87 block L12–L10 interaction. Compound IMB-84 or IMB-87 was incubated with GST-L12 proteins bound to glutathione-sepharose beads, and then His-tagged L10 was added. Equal volume of DMSO was added as control. Protein bands of resulting pull-down products are shown after Western blotting with anti-GST and anti-His antibodies. Data are shown as the mean±SD, n=3. *P<0.05 vs control group.
