GTPVΔ135 is attenuated in inhibition of NF-κB activation. HEK293T cells were infected with AV41 or the Δ135 virus at MOIs of 0.01, 0.1, and 1. Twenty-four hours after infection, cells were cotransfected with pNFκB-luc and pTK-Renilla. At 24 h posttransfection, cells were treated with 100 ng/ml of TNF-α or left untreated for 8 h. Cell lysates were prepared for assays of NF-κB-inducible luciferase activity (A), and mRNA was extracted for determination of the IL-1β mRNA level by qPCR (B).
