Organization of the vTF complex. (A) pM87 interacts with pM92. The FLAG-tagged pM87 expression plasmid or the empty vector was cotransfected with the indicated HA- or Myc-tagged vTFs into HEK293T cells. Cell lysates were immunoprecipitated by anti-FLAG M2 beads and assayed by immunoblotting. (B) Pairwise interaction screening of five vTFs via coimmunoprecipitation. The experiments were performed as described in the legend to panel A, except that the indicated plasmids were used in the samples. (C) Additional interactions were established after coexpression. The experiments were performed as described in the legend to panel A, except that various combinations of vTF subunits, as indicated, were coexpressed in the samples. exp., expression. (D) Model of the MCMV vTF complex. Three pairs of vTF subunits (pM87-pM95, pM95-pM49, and pM91-pM79) form strong interactions, as indicated by black lines. Two of them (pM49-pM95 and pM91-pM79) bind strongly with each other to form a stable subcomplex. The subcomplex binds to pM87 through multiple interfaces. pM92 weakly binds to the complex by interacting with pM87.
