2-ME effect on protein synthesis is decreased in cells expressing PKRmutant protein. MG63 osteosarcoma cells expressing vector (pcDNA) and PKRmutant protein were treated with vehicle and 2-ME for 24 h. The cytoplasmic extracts prepared were subjected to cap-binding (A) or IP with anti-4E-BP1 antibodies (B) and analyzed by western blot analysis using anti-eIF4E antibodies.
